摘要
目的 观察了红花活血化瘀有效成分羟基红花黄色素A(HSYA)抑制血小板活化因子 (PAF)诱发的家兔血小板粘附、5 -羟色胺 (5 HT)释放及血小板内游离Ca2 + 浓度升高的作用。方法 荧光分光光度法检测PAF激活血小板释放的 5 HT ,Fura 2荧光分光光度法观察PAF诱发家兔血小板内游离Ca2 + 浓度的升高及以分光光度法检测PAF所致家兔血小板粘附性的升高。结果 4 79 6~1918 3μmol/L的HSYA抑制PAF诱发的家兔血小板粘附呈明显的量效关系 ;2 80~ 132 0 μmol/L的HSYA抑制PAF诱发的家兔血小板 5 HT释放呈明显的量效关系 ;6 2 9~ 2 0 1 2 μmol/L的HSYA抑制PAF诱发的家兔血小板内游离Ca2 + 浓度的升高呈明显的量效关系。结论 HSYA可明显抑制PAF诱发的家兔血小板粘附、5 HT释放及血小板内游离Ca2 +
Objective To observe the inhibitory effects of Hydroxysafflor Yellow A (HSYA), which is an effective component of the dried flowers of Carthamus tinctorius used for activating blood and resolving blood stasis in TCM, on platelet activating factor (PAF)-induced rabbit platelet adhesion, 5-hydroxy tryptamine (5-HT) release, and increase in the level of free intraplatelet Ca 2+ of rabbit platelets. Methods Fluorospectrophotometry was used to detect the 5-HT released by the rabbit platelets activated by PAF, Fura-2 fluorospectrophotometry was used to observe the PAF-induced increase in the level of free intraplatelet Ca 2+ of rabbit platelets, and spectrophotometry was used to detect the PAF-induced enhancement of adhesion of rabbit platelets. Results The inhibitory effects of HSYA showed a marked dose-effect relationship when HSYA was used to treat PAF-induced enhancement of adhesion of rabbit platelets at doses of 479.6-1918.3 μmol/L, PAF-induced 5-HT release of rabbit platelets at doses of 280-1320 μmol/L, and PAF-induced increase in the level of free intraplatelet Ca 2+ of rabbit platelets at doses of 6.29-201.2 μmol/L. Conclusion HSYA can markedly inhibit PAF-induced enhancement of adhesion of rabbit platelets, PAF-induced 5-HT release of rabbit platelets, and PAF-induced increase in the level of free intraplatelet Ca 2+ of rabbit platelets.
