高效液相法测定银黄胶囊中绿原酸及黄芩苷的含量 被引量:18

摘要 目的 建立高效液相法测定银黄胶囊中绿原酸 ,黄芩苷的含量。方法 Nova pakC18( 2 5 0mm× 4 .6mm ,10 μm)色谱柱 ,绿原酸流动相为乙腈 0 .4 %磷酸 ( 13∶87) ,流速为 1.0mL·min-1,检测波长为 32 7nm ;黄芩苷流动相为甲醇 0 .4 %磷酸 ( 5 0∶5 0 ) ,流速为 1.0mL·min-1,检测波长为 2 80nm。结果 绿原酸在 0 .2 30 1~ 1.6 10 7μg范围内呈良好的线性关系 ,回归方程为Y =2 5 95 .8Х + 1.2 96 4 (r =0 .9999) ,平均回收率为 99.5 8% ,RSD为 1.10 % ,重现性RSD为 0 .4 6 % ;黄芩苷在 0 .2 4 5 3~ 1.4 718μg范围内呈良好的线性关系 ,回归方程为Y =30 4 5 .3Х - 1.3333(r =0 .9999) ,平均回收率为 99.88% ,RSD为 0 .89% ,重现性RSD为 0 .5 6 %。结论 本法简便 ,快速 ,灵敏 ,准确 ,重现性好 。 OBJECTIVE To establish a RP HPLC method for determination of chlorogenic acid and baicalin in Yinhuang capsules. METHOD The Nova pakC 18 (250mm×4.6mm,10mm) column was used .The mobile phase for analysis chlorogenic acid was consisted of acetonitile 0.4% phosphoric acid (13∶87),the detection wavelength was at 327nm. The mobile phase for baicalin was consisted of methanol 0.4%phosphoric acid(50∶50), the detection wavelength was at 280nm. Both of the flow rates were 1.0mL·min -1 .RESULTS The calibration curve of chlorogenic acid was in linear over the range of 0.2301~1.6107μg,and regression equation was U=2595.8Х+1.2964 ( r = 0.999 9 ).The average recovery was 99.58%( RSD =1.10%, n =5), RSD of reproduction was 0.46%. The calibration curve of baicalin was in linear over the range of 0.2453~1.4718μg,and regression equation was U=3045.3Х-1.3333 (r=0.9999).The average recovery was 99.88%( RSD =0.89%, n =5). RSD of reproduction was 0.56%.CONCLUSION The method is simple ,quick ,sensitive ,accurate ,reproducible and can be use to control the quality of Yinhuang capsules.
