摘要
目的 研究血管内皮生长因子C(vascularendothelialgrowthfactorC ,VEGF_C)在大鼠角膜碱烧伤中的表达状况 ,探讨碱烧伤后角膜新生淋巴管形成的分子机制。方法 制作角膜碱烧伤模型 ,运用免疫组化法检测伤后第 1、3、5、7、9天角膜组织中VEGF_C蛋白的表达 ,逆转录_聚合酶链反应 (reversetranscriptionpolymerasechainreaction ,RT_PCR)技术观测伤后第 5天角膜中VEGF_CmRNA的变化 ,电镜观察伤后第 7、14天角膜新生淋巴管状况。结果 碱烧伤后 ,角膜中VEGF_C水平明显升高 ,伤后第 14天角膜出现新生淋巴管。结论 角膜碱烧伤后有新生淋巴管生成 ,VEGF_C在伤后角膜中的高表达可能是角膜新生淋巴管形成的重要机制。
Objective To study the expression of VEGF_C in corneas and the molecular mechanism of lymphangiogensis after corneal alkali burn in rats.Methods The corneal alkali burn animal models were made.Then examined the expression of VEGF_C protein by immunohistochemistry on the 1 st,3rd,5th,7th,9th day after the injury respectively and quantited the level of VEGF_C mRNA with the RT-PCR technology on the 5th day after injury. In addition,the presence of lymphatic vessels in the rat injuried corneas was examined with electron microscope.Resuts The VEGF-C expression in the corneas of rats was dramatically up_regulated after the injury,and on the 14th day after the injury, lymphatic vessels were present in the corneas.Conclusion VEGF_C may be one of an important relevant endogenous factors in corneal lymphangiogensis after alkali burn.
