移植视网膜NOS阳性神经元的发育及其与上丘的联系

摘要 目的:观察不同年龄组段大鼠正常视网膜及移植视网膜内NOS阳性神经元的发育情况及其定位分布,以及移植视网膜能否与视觉中枢建立功能联系。方法:应用NADPH脱氢酶组织化学方法和c-fos免疫组织化学方法显示。结果:(1)正常大鼠视网膜NOS阳性神经元最早出现于生后第5天,第18d数量达到高峰。(2)移植视网膜具有正常视网膜的各层结构和相似的生长发育规律,移植视网膜NOS阳性神经元在生后第4d出现,第12d数量达到高峰,第22d后降至正常成年鼠水平。(3)移植视网膜接受光刺激后,一侧或双侧上丘浅层出现明显的c-fos免疫反应阳性神经元。结论:根据NOS阳性神经元的定位、分布,推测其为无长突细胞、移位无长突细胞及节细胞;移植视网膜能与宿主视觉中枢上丘建立功能联系。 Objective : To investigate the development and distribution of nitric oxide synthase (NOS) positive neurons in the normal and transplanted retina in rats of different ages, and to study whether the transplanted retina has functional connection with the visual center or not. Methods : The NADPH histochemistry and c-fos immunohistochemistry methods were used to show the positive cells. Results : (1) NOS positive neurons in the normal retina appeared 5 d after birth and peaked after 18 d. (2) The intracranially transplanted retina had the same structure and growth regularity as those of the normal retina. The NOS positive neurons in the transplanted retina appeared 4 d after birth, and peaked 12 d after birth and decreased to the normal level after 22 d. (3) The c-fos expressed in one or both sides of the superior colliculus after flash stimulation in the transplanted retina. Conclusion : According to the distribution and the shape of the NOS positive neurons in the transplanted retina, the cells are amacrine cells, displaced amacrine cells and ganglion cells. The transplant ed retina has functional connection with the visual center of the host.
