摘要
目的:观察实验性变态反应性脑脊髓炎(EAE)大鼠丛上细胞的变化。方法:以豚鼠脊髓匀浆和完全福氏佐剂制备急性EAE大鼠模型,以扫描电镜观察各实验组丛上细胞的变化。结果:正常对照组丛上细胞较少,胞体扁平或长杆状,伪足体积大、数目少,放射状分布于胞体周围。EAE发病前组丛上细胞较正常对照组增多,胞体较丰满,伪足多且分布规律,丝状伪足和扁铲样结构分别位于胞体的两侧。EAE发病组丛上细胞显著增多,胞体呈球形,表面可见泡球样结构,伪足以丝状为主,数量少且分布不规律。结论:EAE发病前就有循环中的免疫细胞从脉络丛进入CSF,丛上细胞以巨噬细胞为主,可能与EAE的脑室周病变有关。
Objective : To observe the changes of epiplexus cells in rats with experimental allergic encephalomyelitis. Methods : Rats were actively immunized with guinea pig spinal cord homogenate(GPSCH)-complete freund adjuvant (CFA) emulsions to induce acute EAE. Scanning electronic microscopy was used to observe the changes of epiplexus cells of rats in 3 groups: normal control group, pre-episode group and during episode group. Results : Epiplexus cells were few in normal control group; their soma were flat or like a rod with several large radiate pseudopodia. In pre-episode group, the number of epiplexus cells increased; their soma became greater, pseudopodia distributed regularly: filopodia and flat spade-like structure distributed at amphi-soma. In the episode group, the number of epiplexus cells increased markedly; their soma were in round shapes with many bubble-like structures on the surface; most of the pseudopodia were filament-like, which was few and distributed irregularly. Conclusion : Circular immunocytes enter CSF through choroid plexus before episode. Most of epiplexus cells belong to macrophages, and might contribute to circumventricular lesions during EAE.
