摘要 目的:观察血管内皮生长因子(VEGF)及其受体KDR在成熟期正常SD大鼠肺内的表达情况。方法:取18只成熟期SD大鼠,用逆转录-聚合酶链反应(RT-PCR)和改良的SABC免疫组化法检测其肺内VEGFmRNA、VEGF蛋白及其受体2(KDR,激酶插入区包含受体)表达水平。结果:12周龄大鼠肺内有一定量VEGFmRNA(VEGF189 2.13±0.81;VEGF165 0.69±0.22;VEGF121 0.60±0.30)和KDR表达;16周大鼠肺部VEGF mRNA表达较12周龄鼠有所增加,但差异无显著性。20周龄大鼠肺部VEGF165与VEGF120 mRNA与16周龄对比均明显减少(P<0.05;P<0.01),肺泡上皮及支气管上皮细胞VEGF蛋白表达水平亦较16周龄大鼠显著降低(P<0.01;P<0.01)。结论:VEGF对大鼠肺的生长发育和维持其正常生理功能可能具有一定意义。 Objective;To detect the expression of vascular endothelial growth factor(VEGF) and its receptor KDR in lung tissue of mature stage rat. Methods; 18 12-week old male mature stage SD rats were enrolled in experiment. The expression of VEGF mRNA and VEGF protein and KDR protein in the lung of rats were determined by RT-PCR and modified SABC immunohistochemistry assay separately. Results; There were some VEGF mRNA(VEGF189 2. 13 ±0.81;VEGF165 0.69 ±0. 22;VEGF121 0. 60 ±0. 30) and KDR expression in 12-week rat lung tissue. VEGF mRNA increased in 16-week old rat lung tissue, but had no significant difference (P>0.05). VEGF165 mRNA, VEGF189 mRNA in 20-week rat lung and VEGF protein in alveolar and bronchial epithelium of 20-week rat all decreased(P<0. 05;P<0. 01, P<0.01, P<0.01). Conclusion; VEGF may have a role in the development and normal physiological function in rat lung.
