摘要
目的:探讨急性心肌缺血对大鼠心脏组织内生型一氧化氮合酶(eNOS)基因表达的影响。方法:26只健康成年SD大鼠随机分为对照组(假手术组)、缺血组两组。缺血组取1 h,2 h,24 h三个不同时间点。采用开胸结扎冠状动脉左前降支的方法建立心肌缺血模型,用逆转录聚合酶链式反应(RT-PCR)检测大鼠心梗后不同时间eNOS mRNA表达。结果:冠脉结扎后1 h,两组大鼠缺血心肌组织中eNOS mRNA表达无明显变化(P>0.05);结扎2h时则检测到缺血组大鼠梗死及梗死周围区域心肌组织eNOS mRNA表达下降(P<0.05),eNOS mRNA表达降低持续至结扎后24 h;结扎后24 h组eNOS mRNA表达与结扎后2 h组相比差异无显著性(P>0.05)。结论:在心梗早期(缺血后2 h开始)缺血心脏即有eNOs基因表达减少。
Objective;To observe the expression of endothelial nitric oxide sythase(eNOS) gene in myocardial infarction rats. Methods; twenty-six healthy SD rats were randomly divided into two groups ( controls and ischemia group). Myocardial ischemia was induced by left coronary artery ligation. The eNOS gene expression in ischemic myocardium was assessed using reverse transcriptase-polymerase chain reaction ( RT-PCR) standardized with β-action. Results; ligating of LAD for 2 h and 24 h decreased significantly the eNOS mRNA levels in ischemic myocardial tissue comparing with that in controls (P <0. 05). Conclusion; The results suggested that decreased expression of eNOS gene was one of the most important mechanisms to injure ischemic myocardium in AMI.
