摘要
目的 观测大鼠脊髓损伤后一定时段脊髓组织内 [Ca2 + ]i 的动态变化及亚低温对其的影响。方法 应用激光共聚焦显微镜观测亚低温对大鼠脊髓损伤后 [Ca2 + ]i的影响 ,并与钙组化染色相对照。结果 大鼠脊髓损伤后 [Ca2 + ]i 明显升高 ,至伤后 8h达高峰 ,伤后立即 (5min)亚低温治疗明显减轻细胞内钙聚现象 ,但与对照组相比 ,不能完全阻止 [Ca2 + ]i 的上升。结论 [Ca2 + ]i 升高是脊髓继发损伤病理机制中的重要因素 ,亚低温减轻 [Ca2 + ]i 升高是其脊髓保护机制之一。钙组化染色结果支持上述论点 ,但很难对 [Ca2 + ]i 进行准确快速的动态检测。
Objective To observe the effects of moderate hypothermia on [Ca 2+ ] i after traumatic injury of rat spinal cord. Methods Rats were sacrificed at the l hour, 2 hour, 4 hour, 8 hour, 12 hour after surgery, the calcium level of rats injured spinal cord was respectively measured by calcium histochemical stain and confocal laser-scanning microscopes (CLSM).Results The flourescene intensity of intracellular free Ca 2+ showed a marked increase at l hour that peaked at 8 hour after traumatic injury of rat spinal cord, then maintained a rather high level. Giving moderate hypothermia treatment immediately after injury of rat spinal cord can decrease the increasing of intracellular free Ca 2+ . Conclusions The flouresecene intensity of intracellular free Ca 2+ is an important factors causing secondary injury. Moderate hypothermia treatment can decrease the increasing of intracellular free Ca 2+ , it is one of the protective mechanic of spinal cord. But the dynamic detecting of intracellular free Ca 2+ is difficult.
