摘要 目的 探讨肝细胞生长因子(HGF)对高糖诱导肾间质成纤维细胞损伤的修复作用。方法 将人肾脏间质成纤维细胞培养于不同浓度的葡萄糖中,其中25 mmol/L葡萄糖组培养液中加入不同浓度的rhHGF,分别于不同时间段收集细胞,采用MTT法检测细胞增殖,流式细胞仪检测细胞周期。结果 高糖作用早期细胞增殖旺盛,随着高糖的持续刺激,细胞增殖发生停滞,同时细胞出现肥大。外源性加入HGF后,对高糖造成的细胞损伤具有修复作用。结论 高糖作用下的肾间质成纤维细胞早期出现自限性增殖后细胞增殖停滞,出现肥大。HGF可以通过促进细胞增殖,抑制由于高糖诱导的细胞肥大。 Objective To investigate the damage caused by high glucose, and to measure the role of hepatocyte growth factor(HGF) on fibroblasts. Methods Results Fibroblasts from the human kidney tumor were cul-tured. The cellular proliferation in media containing high glucose and normal glucose was assessed respectively using MTT. Flowcytometry was used to study the cell cycle. The role of exogenous HGF on cellular proliferation was also measured. Results After 36h , in comparison to the control group, high glucose concentration induced the change of cell cycle in fibroblasts. The cells were arrested in the G1 -phase of the cell cycle. At the same time high glucose in-hibited cellular proliferation. HGF modulated the proliferation of fibroblasts. Conclusion In vitro high glucose suppresses human fibroblasts proliferation and induces cellular hypertrophy. Exogenous HGF can prevent cell hyper-trophy.
