用Fura-2/AM测定益康唑诱导鼠白血病WEHI-3细胞凋亡后细胞质游离钙浓度

摘要 目的 通过体外实验 ,研究益康唑 (Econazole)是否可触发鼠白血病细胞株WEHI 3细胞胞质 [Ca2 + ]i浓度的改变。方法 用Ca2 + 荧光探针 (Fura 2 /AM )负载WEHI 3细胞后测定Econazole作用WEHI 3细胞 12、 18、2 4h的胞内 [Ca2 + ]i浓度。结果 Econazole作用WEHI 3细胞 12、 18、 2 4h后的胞内 [Ca2 + ]i均高于对照组(80 6± 15 3)nmol/L ,分别为 (131 2± 11 2 ) ,(15 6 5± 2 0 1) ,(182 1± 13 6 )nmol/L。结论 提示Econazole作用WEHI 3细胞可增加细胞质内 [Ca2 + ]i浓度。 Objective To investigate whether Econazole can trigger the changes in the concentration of cytoplasmic free calcium in mouse leukemia cell WEHI-3 in vitro. Methods WEHI-3 cells were loaded with Fura-2/AM and the concentrations of cytoplasmic free calcium were determined after the cells were treated with Econazole for 12, 18, 24 h respectively.Results The concentrations of cytoplasmic free calcium in the WEHI-3 cells treated with Econazole for 12, 18, 24 h were all increased as compared with control group \[(131.2±11.2), (156.5±20.1), (182.1±13.6) nmo l/L vs (80.6±15.3) nmol/L\]. Conclusion The concentration of cytoplasmic free calcium in WEHI-3 cells treated with Econazole could be increased.
