摘要
目的 研究SARS病毒合成肽疫苗。方法 根据目前已知SARS病毒的基因组序列及其编码的蛋白质 ,选择了SARS病毒编码的spike蛋白抗原表位蛋白质序列 ,运用计算机抗原表位预测技术 ,筛选抗原表位 ,以合成肽抗原表位。通过对SARS病人血清进行筛选确定与血清高交叉反应的 3个短肽 ,再合成多重抗原肽 (multipleantigenicpeptide ,MAP) ,与KLH偶联后免疫小鼠。结果 筛选的 3条与SARS病人血清反应强的肽 ,免疫小鼠均产生了高效价的抗体。结论 以合成肽为免疫原研究SARS病毒肽疫苗有着广阔前景。
Objective To study the immunogenicity of multiple antigenic peptides (MAP) derived from spike protein of SARS-CoV as a preliminary approach toward formulating a vaccine. Methods Computer algorithm was applied to select dominant epitopes from the published sequence of spike protein of SARS-CoV. Eight peptide sequences varying from 15-mers to 39-mers were synthesized and then tested by convalescent sera. Three of them showed eviden reactivity with convalescent sera(40/46;27/46;24/46) were synthesized as branched pedtides,linked to KLH and were used to immunize mice to test it′s immunogenicity. Results Synthetic peptide antigens can induce high titer of specific antibodies. Intramuscular route was more efficient than nasal instillation. Conclusion Synthetic peptide as immunogens for SARS-CoV seems to have a good prospect.
