安体优抑制小鼠Lewis肺癌血管生长作用的实验研究 被引量:5

摘要 目的 :探讨安体优抑制小鼠Lewis肺癌血管生长的作用及其抗Lewis肺癌转移的相关机理。方法 :利用Lewis肺癌细胞肺转移模型 ,随机分为三组 ,分别予安体优、蒸馏水、环磷酰胺治疗 ,运用免疫组化方法对移植瘤中血管内皮细胞生长因子(VEGF)的表达、微血管密度 (MVD)等有关转移指标进行检测和分析。结果 :安体优组移植瘤血管内皮细胞生长因子 (VEGF)的高表达率 30 %和微血管密度 (MVD) 1 4 .3± 5 .38,空白对照组VEGF的高表达率 80 %与MVD1 9.8± 3 .65 ,经统计学处理有显著性差异VEGF高表达率与MVD的关系经直线相关分析 ,呈显著正相关 (P <0 0 5)。结论 安优优可以降低移植瘤MVD与VEGF高表达率 。 Objective:To explore the mechanisms of ANTIYOU on depressing Lewis lung carcinoma vascular growth of C 57 BL/6 mouse for supplying the experimental evidences for clinics.Methods: Lewis lung carcinoma cells(5×10 5/mouse) were inoculated subcutaneously to 60 mice in right axillas, they were randomized into 3 groups after 48 hours.The treated groups were given ANTIYOU and CTX respectively and the control group was only given distilled water.Ten mice of each group were killed on the 22nd day. Vascular endothelial growth factor (VEGF)protein and microvessed demstity(MVD) expression in all 30 subcutaneous tumors were detected by immunohistochemistry method.Results: The high expression ratios of VEGF and in each of ANTIYOU and CTX group are lower than those in control group significantly ( P <0 05, P <0 01 respectively).Conclusion: Making molecules VEGF and MVD less expression in carcinoma cells is one of possible mechanisms to anti metastasis to lungs in C 57 BL/6 mouse with Lewis lung carcinoma.MVD in the tumors of control group was significantly higher than in the ANTIYOU group,and positively correlated with the VEGF expression.
