摘要
背景与目的:胶质瘤增殖动力学是其重要生物学特征之一。本文研究探讨大鼠体内C6胶质瘤的增殖动力学特点。方法:建立C6/Wistar胶质瘤大鼠模型,取肿瘤组织分别行流式细胞仪测定,AgNOR染色和PCNA免疫组化染色。结果:C6胶质瘤细胞S期百分比为18.19%,PCNA指数为0.77,AgNOR计数和面积分别为3.2个/细胞核、10.67μm^2/细胞核。三组指标均具有相关性(P<0.05)。结论:C6胶质瘤细胞是一种具有高度增殖活性的恶性肿瘤细胞,应用流式细胞仪检测、PCNA免疫组化染色及AgNOR染色三种方法可以从不同角度反映肿瘤细胞增殖状况,为进一步客观评价治疗因素对胶质瘤细胞增殖活性的影响及判断预后奠定实验基础。
BACKGROUND & OBJECTIVE: Proliferative kinetics of glioma is one of the most important biology characteristic. This study was undertaken to investigate the proliferative kinetics of C6 glioma in rat. METHODS: After establishing C6/Wistar brain glioma model, we carried out flow cytometric analysis, PCNA immunohistochemistry and AgNOR stain. RESULTS: S phase fraction of C6 glioma cell was 18.19%. PCNA index was 0.77. AgNOR number and area were 3.2/cell nucleus, 10.67μm^2/cell nucleus. All of the indexes in three methods had statistical correlation(P<0.05). CONCLUSION: C6 glioma cells are highly proliferative tumor cells. By using flow cytometric analysis, PCNA immunohistochemistry and AgNOR stain, the proliferative kinetics can be reflected in different aspects. These findings provided the foundation for objective evaluation of the effects of therapeutic agents in proliferative kinetics and in determining prognosis.
