摘要
中国弯颈霉Tolypocladium sincnse Li与雪白弯颈霉Tplypocladium niveum(Rostrup)Bisset的孢子悬浮液经γ-射线Co^(60)辐射诱变后,产环孢菌素的得率及抑菌活性均明显提高.用黑曲霉Aspcrgillus niger和弯孢霉Curvularia lunata作为致敏菌,检测提取物中环孢菌素的抗菌活性,提取物抑制了致敏菌的菌丝生长和孢子形成.环孢菌素产生菌深层发酵培养基是蛋白胨—葡萄糖培养基,发酵周期12天.尔后发酵液用等体积的乙酸乙酯萃取,减压浓缩,每升发酵液获环孢菌素粗制品35—87mg.
The suspension of spores of Tolypocladium sinense Li and Tolypocladium nivcum (Rostrup) Bisset was irradiated by different doses of y-ray Co^(60) in this experiment. After induced variations, the yield of cyclosporin was obviously increased. The Aspergillus nigcr and Curvularia lunata were used to detect antifungal activity produced by a crude extract of cyclosporin which can inhibit the growth of hyphae and the formation of eonidia. Two irradiated strains were submerged in culture medium liquor of peptone-glucose, at 26℃, for a fermentation period of 12 days. The fermentation broth was extracted with an equal volume of acetic ether. After separation the organic layer was evaporated under reduced pressure from 1L of fermentation broth, extracted. In this way about 35-87 mg of crude matcral were obtained
