摘要
目的 探讨灵芝多糖 (GLP)对顺铂 (CDDP)所致肾毒性的预防作用和对其抑瘤率的影响。方法 雌性Wistar大鼠接种W2 56癌肉瘤后随机分为生理盐水组、灵芝多糖组、顺铂组、顺铂 +灵芝多糖组 ,连续给药 10d后采样 ,分别测血肌酐 (Scr)、尿素氮 (BUN)、肾皮质组织丙二醛 (MDA)含量、超氧化物歧化酶 (SOD)活性、谷胱甘肽过氧化物酶 (GSH -Px)活性 ,并计算抑瘤率。结果 顺铂组血清Scr、BUN明显升高 ,肾皮质组织MDA含量升高 ,SOD与GSH -Px活性降低 ,与生理盐水组比较有显著差异 (P <0 0 5 ) ;顺铂 +灵芝多糖组血清Scr、BUN降低 ,肾皮质组织MDA含量降低 ,SOD活性、GSH -Px活性升高 ,与顺铂组比较有显著差异 (P <0 0 5 ) ;顺铂 +灵芝多糖组抑瘤率与顺铂组比较无显著差异 (P >0 .0 5 ) ,与灵芝多糖组、生理盐水组比较有显著差异 (P <0 .0 5 )。结论 灵芝多糖能减轻顺铂肾毒性 ,其机制可能与抑制顺铂致肾皮质脂质过氧化增强有关 。
Objective To explore the preventive actions of ganoderma lucidum polysaccharide on nephrotoxicity induced by cisplatin and the influence on carcinoma inhibition rate.Methods Female Wistar rats were randomly divided into NS group,GLP group,CDDP group,and GLP+CDDP group after being infected with W 256 carcinoma celles.The changes of Scr,BUN,MDA,SOD,and GSH_Px of renocortical tissue were measured and carcinoma inhibition rate was calcuated after 10 days by injecting drugs.Results The canter of Scr and BUN in CDDP group was significantly higher than of NS group.The content of MDA increased and the activity of SOD and GSH_Px declined in renocortical tissue(P<0.05).The content of Scr and BUN in GLP+CDDP group was significantly lower than that of CDDP group(P<0.05).The concent of MDA declined and the activity of SOD and GSH_Px increased in renocortical tissue(P<0.05).There was no significant difference on carcinoma inhibition rate between GLP+CDDP and CDDP group(P<0.05).Conclusion GLP may reduce cisplatin nephrotoxicity and its mechanism may be correlative with that GLP inhibited renocortical tissue lipid peroxiction reaction increasing.GLP does not influence cisplatin activity in anti_tumor.
