摘要
应用核糖体 DNA ITS区的 RFLP分析对 3 8个不同寄主来源的弯孢类炭疽菌菌株的遗传多样性和系统发育进行研究。结果表明 :弯孢类炭疽菌的 ITS扩增区 ( ITS4 ~ITS5)约为 65 0 bp,无长度变异。 5种内切酶 ( Alu I、Bsu RI、H in6I、HpaII和 Taq I)的酶切图谱在种内是相似或一致的 ,而种间差异较大。ITS-RFLP共迁带 UPGMA聚类分析的结果表明 :3 8个弯孢类炭疽菌菌株被聚为 6个类群 ,群与群之间分界明显 ,表明种的分界相当明显。ITS-RFLP分析的结果还揭示了一些近似种的分类关系 ,如 ,按传统方法分别鉴定为 Colletotrichum truncatum、C.circinans和 C.capsici的许多菌株有相似或完全一致的内切酶酶切图谱 。
Genetic divergence among 38 strains belonging to the falcate spored species of Colletotrichum was assessed by RFLP analysis on the basis of rDNA ITS region. The PCR amplified ITS region (ITS 4-ITS 5) was about 650 bp in length in all the tested strains. RFLP patterns of ITS products digested with different endonucleases ( Alu I, Bsu R I, Hin 6 I, Hpa II and Taq I) were not distinguishable within same species, but clearly different at the interspecies level. UPGMA analysis of co migrate band in restriction patterns showed that 38 isolates could be divided into 6 distinct groups. Some strains previously in the different species, such as C. truncatum, C. circinans and C. capsici , were closely grouped together on a cluster dengrogram, indicating that they were possible belong to the same species.
