摘要
对柿品种‘禅寺丸’(DiospyroskakiThunb .cv .Zenjimaru)花粉进行了干燥脱水法 (Dehydration)和玻璃化法 (Vitrification)液氮超低温保存研究 ,结果表明 :0℃下硅胶干燥 8h或 (2 0± 1)℃下玻璃化液PVS2 处理4 0~ 6 0min后直接投入液氮保存 ,4 0℃解冻 70s后 ,相对存活率 (TTC检测 )和萌发率都在 90 %以上 ,且活力与保存时间长短无关。该结果为柿花粉的长期保存提供了技术支持。
Researches have been carried out on cryopreservation of pollen of 'Zenjimaru' persimmon(Diospyros kaki Thunb.). The pollen was r apidly plunged into liquid nitrogen for storage following dehydrating for 8 h wi th silica gel at 0 ℃ or incubating in PVS 2 for 40~60 min at (20±1) ℃ . After rapid thawing for 70 s in water at 40 ℃,both of the relatively sur vival rate(tested by TTC)and germination acpacity exceeded 90%, and the viabi lity was not influenced by the duration of storage in liquid nitrogen.This metho d provided a simple strategy to conserve the persimmon pollen for a long time.
