摘要
病毒性出血症病兔肝悬液离心去杂后其上清用聚乙二醇浓缩,又经正丁醇-异戊醇除去杂蛋白,然后以差速离心和蔗糖梯度离心,提取兔出血症病毒(RHDV)。提纯的病毒用核酸酶降解污染的细胞核酸后,加入8mol/L尿素,使核酸从病毒粒子中释放出来,经水相法展开,附于有碳膜的铜网上。检样经醋酸铀染色和铂旋转喷涂投影,于透射电镜下观察。RHDV核酸呈线状,平均长度为1.92μm。经公式推算出分子量为2.30×10~6道尔顿(Dalton)。
Rabbit haemorrhagic disease virus (RHDV) was purified from the infected rabbit livers by using the mixture of n-butanol and isoamyl alcohol, combined with sucrose density gradient centrifugation. DNase I and RNase A were used to eliminate the contaminated nucleic acid derived from host cells. After identified by HA test and electronmicroscopy, the purified RHDV was treated with 8mol/L urea and the released viral DNA was transferred tothe electron microscope grids by aqueous technique. The specimens stained with uradium acetate and shadowed with platinum were examined under electron microscope. The molecules of RHDV-DNA were found to be linear with average length of 1.92μm. The molecular weightof RHDV-DNA was calculated to be 2.30×10~6 daltons.
