摘要
目的建立高效液相色谱法测定蛹虫草人工固体培养菌皮中核苷类化合物的含量。方法采用pH 6 .5磷酸盐缓冲液 甲醇 (85∶15 )为流动相 ,流速为 1.0ml/min ,检测波长为 2 6 0nm。结果蛹虫草人工固体培养物菌皮中主要成分虫草素在 4 .0 3~ 10 0 .8μg/ml范围内线性关系良好 ,回归方程为A =39.5 8C - 0 .0 342 ;尿苷在 3.6 8~ 92 .0μg/ml范围内线性关系良好 ,回归方程为A =2 4 .32 9C - 0 .0 0 2 7;腺苷在 3.84~ 96 .11μg/ml范围内线性关系良好 ,回归方程为A =18.0 12C - 0 .0 2 35。结论人工固体培养菌皮中核苷类成分的含量随培养时间发生一定的变化 ,高效液相色谱法可用于蛹虫草人工培养物中核苷类化合物的测定 ,为控制其质量提供了简便易行的方法。
PurposeTo develop a method for the determination of nucleosides in lawn of cultured Cordyceps militaris by HPLC.MethodsThe contents of nucleosides were determined using HPLC on Hypersil C 18 column with pH 6.0 phosphate-methnol(85∶15) as mobil phase and detected at 260 nm. The running rate was 1.0 ml/min.ResultsThe linear ranges of cordycepin, adenosine and uridine were got in the concentration range of 4.03~100.8, 3.84~96.11, 3.68~92.0 μg/ml,respectively.The regression equations of them were A=39.58C-0.034 2, A=18.012C-0.023 5, A=24.329C-0.002 7, respectively.ConclusionThe results showed that this method can be used for the analysis of cultured Cordyceps militaris.
