摘要
目的 :探讨单次光化学疗法 (photodynamictherapy ,PDT)治疗小鼠皮下种植E7+ U14肿瘤的疗效 ,为临床PDT治疗相关肿瘤提供实验依据。方法 :根据肿瘤体积决定光源输出的光斑面积和计算光照射时间 ,比较PDT治疗组与荷瘤对照组的肿瘤生物学特点及荷瘤小鼠的生存期。结果 :所有小鼠瘤细胞均成功种植 ,肿瘤体积呈对数增长 ,在局部呈浸润性生长 ;单次PDT治疗后肿瘤细胞大量坏死 ,肿瘤体积无明显增长 ,且显著小于同期荷瘤组 ;PDT治疗能有效降低肿瘤的肺和淋巴结转移率 ,并将荷瘤小鼠的平均生存期从 2 8.9d延长到 37.6d。结论 :单次光化学疗法通过损伤小鼠皮下种植E7+ U14肿瘤细胞改变瘤细胞的生物学特点 ,并有效延长荷瘤动物的生存期 ,可在相关肿瘤治疗方面进行更为深入的应用研究。
Objective: The simple photodynamic therapy(PDT) was adopted to trea t the implanted U14 carcinoma in 615 mice,to evaluate its therapeutical effect s and provide experimental basis for tumor therapy. Methods: The area of light s pot and the time of irradiation was adjusted according to the tumor volume,biol ogic behavior of the tumor in control group and PDT group and the survival time was observed. Results: All the mice survived after U14 carcinoma implanted,the tumor mass grew logarithmicly and infiltrated into the chest wall,massive nec rosis of U14 carcinoma cells in tumor sphere after PDT,the volume of tumor did not increase dramatically compared with the control group,the tumor metastases rate decreased significantly in the lungs and lymph nodes,the average survival time in mice with U14 carcinoma were prolonged from 28.9 to 37.6 days b y simple PDT. Conclusion: Biologic behavior of the tumor were changed by simple PDT,the average survival time in mice with U14 carcinoma were prolonged,more a pplication investigation in tumor therapy with PDT were needed.
