摘要
目的 建立溃疡性结肠炎大鼠模型。方法 经结肠给大鼠注入 8%乙酸溶液 ,每日观察大鼠一般情况 ,10d后处死大鼠取出结肠 ,肉眼观察并做组织学切片进行镜下观察 ,同时应用免疫组化技术检测结肠黏膜内TNF α、IL 10、NF κB细胞因子的表达。结果 大鼠的一般情况、大体病理及组织学病理均与人类溃疡性结肠炎类似 ,并且结肠黏膜TNF α、NF κB表达明显升高 ,IL 10表达明显降低 ,上述细胞因子变化亦与人类溃疡性结肠炎的变化基本一致。结论 该大鼠模型重复性好 ,方法简单 ,短期内即可出现明显症状 ,适合于研究溃疡性结肠炎的发病机制及临床新药开发 ,同时还可用于评价新药疗效。
Objective To establish the ulcerative colitis (UC) rat model. Methods The rat model of ulcerative colitis was established by colonic perfusion of 8% acetic acid. The symptoms and microscopic observation were recorded, and the expression of tumor necrosis factor(TNF)-α、interleukin(IL)-10 and NF-κB in colonic mucosa were detected by semi-quantative immunohistochemistry. Results Symptoms,colonic histological changes occurred.Levels of TNF-α and NF-κB were increased and level of IL-10 decreased in colonic mucosa (P<0.05). The changes of cytokines were basically the same as human. Conclusion The method is simple and the symptoms occur in a short time. It is well suitable for analyzing the pathogenic mechanism and assessing the effects of a new drug.
