长江口刀鲚遗传多样性的随机扩增多态DNA(RAPD)分析 被引量:32

摘要 用随机扩增多态DNA(RAPD)技术对长江口刀鲚 (Coiliaectenes) 30个个体的遗传多样性进行了检测 ,从 4 0个随机引物中筛选出 17个对每个刀鲚的DNA进行扩增 ,结果表明 ,17个引物共检测到 14 8条清晰且重复性好的条带 ,分子量在 2 0 0~ 2 0 0 0bp之间 ,其中多态位点为 86个 ,占5 8 11% ;群体的Shannon多样性指数为 0 190 5 ,Nei基因多样性指数为 0 2 2 80 ;个体间最大遗传距离为 0 2 12 ,最小遗传距离为 0 0 92。通过与其他鱼类的遗传多样性的研究结果比较可初步判断 ,刀鲚群体的遗传多样性比较丰富。 Random amplified polymorphic DNA (RAPD) technique was employed to detect the DNA polymorphism in 30 individuals of natural population of Coilia ectenes. 17 arbitrary primers were used and 148 clear,repeatable RAPD bands were generated. Each primer used for the detection could produce 4~12 molecular markers and the reproducible fingerprints varying in length from 200~2 000 bp. The percentage of polymorphic fragments was 58.11%, while the index of Shannon diversity was 0.190 5 and the index of gene diversity was 0.228 0. The genetic distance between individuals was from 0.09 2 to 0.212. Comparing the genetic diversity of Coilia ectenes with that of other fishes, the experiment revealed that the genetic diversity of Coilia ectenes was in much higher state. Owing to lack of historic data, we could not judge the effect on its genetic diversity caused by excessive fishing and environment destruction according to the results of this research.
