Extraction and Characterization of Resistance-elicitors from Haphal Cell Wall of the Rice Blast Fungus Magnaporthe grisea

摘要 采用离心、超滤、DEAE SepharoseFF阴离子交换层析柱等方法对稻瘟菌 (Magnaporthegrisea)细胞壁来源的激发子活性物质进行了提取 ;以一套水稻抗稻瘟病近等基因系为材料 ,对其诱导水稻叶片植保素、过氧化物酶 (POD)与苯丙氨酸解氨酶 (PAL)活性进行了检测 ;结果表明 ,粗激发子提取物 (CEF)及DEAE SepharoseFF层析提取物DSⅡ和DSⅢ菌具有激发子活性。经热、胰蛋白酶和过碘酸钠处理后的生物活性检测结果表明DSⅡ和DSⅢ的活性位点均位于糖基部分。 Resistance-elicitors from hyphal cell wall of a strain in the race ZC13 of Magnaporthe grisea was isolated through centrifugation, ultra-filtration and DEAE-Sepharose FF chromatography. Eliciting activities of the extracted fractions were carried out on four near isogenic lines of rice. The results showed that the crude elicitor fraction (CEF), the two purified fractions DSⅡand DSⅢcould increase the levels of the activity of peroxidase (POD) and phenylalanine ammonia-lyase (PAL) in rice leaves. Digestion of DSⅡand DSⅢ using trypsin and boiling at 100 ℃ for 10 min, did not diminish their eliciting activity. A complete loss of the eliciting activity of DSⅡand DSⅢ was observed when the fractions were treated by periodate. The results showed that the carbohydrate residue is probably the active site in DSⅡand DSⅢ, and the protein may be less important in the eliciting activity.
