大鼠心脏移植后外周血T细胞亚群的动态变化 被引量:1

摘要 目的 探讨心脏移植后大鼠T细胞亚群的动态变化与急性排斥反应的关系。方法 用流式细胞仪测定Wistar→SD大鼠心脏移植组和正常SD大鼠组外周血T细胞亚群及比值 ;对心脏移植组急性排斥反应 (AR)进行病理分级。结果 心脏移植组AR在术后第 5天最严重 ,第 7天减轻 ;CD4 + T及CD8+ T比正常大鼠组增高 ;CD4 + T在术后第 3,5天高于正常 ,CD8+ T在术后第 1,5 ,7天出现显著性增高 ;CD4 + T/CD8+ T的比值在术后第 7天低于其他时间段和正常组 ,且比值小于 1。结论 心脏移植后CD4 + T及CD8+ T总体细胞数升高 ;当杀伤性CD8+ T细胞上升时 ,排斥反应有所加重 ;CD4 + T/CD8+ T的比值 ,在心脏移植后第 7天低于 1,同时排斥反应减轻 。 Objective To study the relationship between the changes of T lymphocyte subsets and acute rejection in heterotopic heart transplantation rats. Methods Heart transplantation was performed from Wistar to SD rats. T lymphocyte subsets was measured by flow cytometry and acute rejection was graded according to pathalogical changes. Results The CD4 + T cell and the ratio of CD4 + T/CD8 +T in the experiment group increased on the 3rd day then decreased on the 5th day and 7th day after heart transplantation. The CD8 + T in experiment group decreased on the 3rd day and then rose rapidly on the 5th day and 7th day. In experiment group, the acute rejection was most prominent on the 5th day and improved on the 7th day. Conclusion After heart transplantation CD4 + T cell and the ratio of CD4 + T/CD8 +T increase followed by a rapid decrease, but CD8 + T cell decreases with subsequent increase. Acute rejection is the 5th day with subsequent improvement on 7th day. Acute rejection is most prominent on is attenuated on the 7th day after heart transplantation and the ratio of CD4 + T/CD8 +T is below 1, suggesting that heart transplantion may produce immune tolerance.
