胰岛素对家兔血浆支、芳氨基酸浓度与其比值的影响

摘要 给饥饿家免按每公斤体重4u于皮下注射药用胰岛素。注射前、后30分钟,分别抽血并分离出血浆。用聚酰胺薄层析分离和荧光测试法检测血浆支链氨基酸(BCAA)及芳香族氨基酸(AAA)荧光强度,并以此计算支/芳(BCAA/AAA)比值。以对照液用同样方法做对照组实验。研究结果,AAA变化不显著,但BCAA则明显降低,因而支/芳比值从注射前的2.816下降至注射后的1.486,其下降百分率为48.0%。提示,增加血浆胰岛素的有效浓度能显著改变BCAA的分布平衡。病例测试,选择具有高胰岛素血症的肝硬化患者20例及肝性脑病患者13例,以同样方法进行血浆支/方比值测定,所获结果与上述动物实验的相互吻合。 The concentrations of the BCAA (V. I. L) and AAA (F, Y) were determined and themolar ratio of BCAA/AAA calculated in the plasma of fasting rabbits 30 minutes afterhypodermic insulin (4 I. U./kg. body(?)wt.). Strongly fluorescent dansyl amino acids bythin-layer microchromatography were identified and measured. The experimental groupshowed an increase in plasma BCAA whereas the change in AAA concentrations was nil.The calculated BCAA/AAA ratio decreased from 2.816 to 1.486 after the injection. Theresults suggested that an effective dose of insulin caused a change in the distributionequilibrium of the plasma BCAA. Plasma BCAA/AAA in patients with liver cirrhosis (20 cases) and chronic hepato-encephalopathy (13 cases) showed similar changes.
