摘要
目的 应用荧光定量聚合酶链反应技术验证由基因芯片发现的干扰素 (IFN)诱导基因表达谱在系统性红斑狼疮 (SLE)、类风湿关节炎 (RA)和正常人中的表达。方法 测定 4 0例SLE患者、2 8例RA患者和2 9名正常人外周血白细胞Ly6e、IFIT1、OAS2IFNw、IFNa13表达 ,初步分析外周血分离出来的T、B及单核细胞中这些基因的表达。结果 与正常人相比 ,SLE组的Ly6e、IFIT1、OAS2基因表达显著增高 ,其中Ly6e、OAS2基因的表达较RA组也显著增高 ,但IFNw、IFNa13基因表达反而降低 .这组基因在单核细胞的表达最高。结论 IFN诱导的基因在SLE患者中呈高表达 。
Objective To validate the interferon (IFN)-inducible gene expression spectrum discovered by microarray in systemic lupus erythematosus(SLE), RA patients and normal controls using real-time PCR. Methods Peripheral blood obtained from 40 SLE, 28 RA patients and 29 normal controls had their total RNA extracted and cDNA synthesized. Real-time PCR was used to determine the Ly6e,IFIT1,OAS2, IFNw, IFNa13 expressions at transcription level. The expressions of IFN-inducible genes in T, B and monocytes isolated from normal subjects blood were tested. Results The expressions of Ly6e,IFIT1,OAS2 genes were upregulated in SLE vs normal controls, and the expressions of Ly6e,OAS2 genes were also upregulated in SLE vs RA patients. While the expressions of IFNw, IFNa13 genes were downregulated. The expressions of IFN inducible genes were highest in monocyte subsets of leukocyte. Conclusion The IFN inducible genes are up-regulated in SLE patients, which may play an important role in the pathogenesis of SLE.
