摘要
目的 利用体外培养的兔视网膜M櫣ller细胞 ,观察其在高糖、高胰岛素条件下血管内皮生长因子 (VEGF)表达的变化。 方法 采用免疫细胞化学法、原位杂交法和酶联免疫吸附法(ELISA)进行定性定量测定高糖、高胰岛素条件下体外培养的M櫣ller细胞VEGF的表达?〗峁「咛恰⒏咭鹊核啬艽碳櫣ller细胞VEGF的表达 ,并且是通过转录水平调节的 ,高糖对M櫣ller细胞分泌的VEGF的调节呈时间和剂量依赖性。 结论 高糖和高胰岛素都可以通过刺激M櫣ller细胞编码VEGF基因转录 ,增强VEGF蛋白表达 ,而在糖尿病视网膜病变 (DR)的新生血管形成中发挥重要作用。
Objective To investigate the changes of vascular endothelial growth factor(VEGF) in cultured rabbit retinal Müller cells at high glucose and different insulin concentrations in vitro. Methods Immunocytochemical method, in situ hybridization and ELISA were used to assay the change of VEGF expression in cultured Müller cells in vitro at different i nsulin concentrations in qualitative and quantitative ways. Results VEGF expression at transcription was enhanced markedly by the high level s of glucose and insulin. The effect of high glucose on Müller cells was time- and dose-dependent. Conclusions VEGF expression in cultured Müller cells in vitro can be enhanced at high levels of glucose and insulin. T he increased VEGF expression of Müller cells maybe plays a potential role in ne ovascularization of diabetic retinopathy.
