高同型半胱氨酸血症对糖尿病肾病小鼠细胞间黏附分子-1表达的影响 被引量:6

摘要 目的 探讨高同型半胱氨酸 (Hcy)血症对糖尿病肾病 (DN)时细胞间黏附分子 1(ICAM 1)表达的影响。 方法  2 2~ 2 4周龄Kkay糖尿病鼠 16只随机分两组 :糖尿病 (KA)组和高蛋氨酸 (KB)组 ,每组 8只 ;另以 9只C5 7BL/ 6小鼠为对照 (C5 7) ,以相应饲料喂养 4个月后取材。测定血糖和血Hcy水平 ,以PAS染色观察各组DN病变。对肾脏ICAM 1蛋白表达进行免疫组化染色和半定量图像分析 ;并以RT PCR检测肾脏ICAM 1mRNA的表达水平。 结果 与C5 7对照组相比 ,Kkay两组均有不同程度DN病变 ,其中KB高蛋氨酸组病变最重 ,其肾小球硬化指数 (GI)高于KA组 (P <0 0 5 )。免疫组化显示Kkay小鼠肾脏ICAM 1的表达高低与病理变化一致 ,即KB组ICAM 1阳性着色面积 (19 6 5 % )高于KA组 (15 2 2 % ) (P <0 0 1) ;ICAM 1mRNA表达水平也强于KA组。 结论 Hcy具有加重Kkay小鼠DN程度的作用 ,此作用可能与上调ICAM Objective To study the effect of hyperhomocystein e (Hcy) on diabetic nephropathy of type 2 diabetes and the relation of Hcy with the expression of intercellular adhesion molecule-1(ICAM-1). Metho ds Sixteen Kkay mice were divided into two groups, 8 in each:diabetes group (KA) and methionine-diet group (KB). C57Bl/6 mice were used as normal con trol (C57). At four months after treatment with two different diets, the m ice were sacrificed and serum homocysteine was assayed by fluorescence polarizat ion immunoassay. The renal pathological changes were examined by periodic acid -schiff (PAS) stain. The expression of ICAM-1 protein and mRNA were detec ted by immunohistochemical staining and RT-PCR. Results The pathological fingdings of diabetic nephropathy in KB group were more serious com paired with KA group based on morphological changes. It was showed that inducti on of hyperhomocysteinemia in Kkay mice (KB) by a diet enriched in methionine ag gravated diabetic nephropathy with higher glomerulosclerosis index (GI) (283.33 ) than that in KA group (240.00) (P<0.05). Also the expressions of ICAM- 1 protein and mRNA levels in the glomeruli were also enhanced. Conclus ions The elevated level of Hcy worsens the degree of nephropathy,which may be related to the higher expression of ICAM-1.
