垂体腺瘤的细胞凋亡及其相关基因产物的研究

摘要 目的 :研究人类垂体腺瘤中的细胞凋亡及其相关基因p5 3、Bcl 2、Bax产物。 方法 :采用TUNEL法检测14例患者垂体腺瘤的细胞凋亡 ,同时应用免疫组化技术检测 17例垂体腺瘤的细胞增殖相关抗原 (PCNA)和凋亡相关基因p5 3、Bcl 2、Bax表达的蛋白产物。 结果 :14例垂体腺瘤中 12例检测到凋亡细胞 (85 .71% ) ,其凋亡细胞比例仅占 0 .1%~ 8.0 % ,与肿瘤体积呈负相关 ,与腺瘤细胞增殖活性关系不密切。凋亡相关基因中Bax阳性率高 (92 .31% ) ,表达强 ,p5 3有 2例阳性 ,Bcl 2仅 1例弱阳性。腺瘤细胞Bax表达和细胞增殖活性呈正相关。结论 :人类垂体腺瘤中存在细胞凋亡。垂体腺瘤中Bax高表达 ,Bcl 2 /Bax比值 <1.0 ,这一细胞内的微环境有利于细胞凋亡的发生。人垂体腺瘤细胞Bax表达和细胞增殖活性呈正相关。垂体腺瘤的凋亡活性和其他临床特征(如增殖活性、侵袭性等 )无明显相关性。 Objective: To investigate apoptosis and its related gene p53, Bcl-2, Bax products in pituitary adenoma of human. Methods:Apoptosis was assayed in 14 cases by the method of TdT-mediated bio-dUTP nick end labelling. At the same time, immunohistochemistry method was used in 17 cases to assay the products of PCNA and p53, Bcl-2, Bax. Results:Apoptosis was found in 12 out of 14 cases (85.71%). The proportion of apoptosis cell was 0.1~8.0%, which was negatively related to the tumour volume, but not correlated to the proliferation ability. 92.31% cases were stained for Bax with dyeing greatly, and only 2 cases were stained for p53, 1 case was stained weakly for Bcl-2. Conclusion:Apoptosis exists in pituitary adenoma of human, possibly owing to the more protein of Bax produced. The expression of Bax is positively correlated with the proliferation index of pituitary adenoma cells. There is no relationship between apoptosis and proliferation ability, invasive ability, or other clinic figures of pituitary adenoma of human being.
