摘要
①目的 检测原发性卵巢癌中p15mRNA及其蛋白的表达 ,以探讨该基因在原发性卵巢癌发生发展过程中的作用。②方法 应用逆转录 聚合酶链反应 (RT PCR)以及免疫组化方法 ,对 4 8例原发性卵巢癌以及 15例正常卵巢组织中 p15mRNA及其蛋白的表达进行检测。 ③结果 原发性卵巢癌中 p15mRNA及其蛋白的表达与正常卵巢组织比较差异有显著性 (χ2 =4 .16、5 .11,P <0 .0 5 )。p15mRNA及其蛋白表达与卵巢癌组织学类型相关 (χ2 =5 .5 9、7.30 ,P <0 .0 5 ) ,与其FIGO分期及组织学分级无关 (χ2 =1.13~ 2 .0 9,P >0 .0 5 )。④结论 原发性卵巢癌中p15mRNA及其蛋白表达异常 ,提示该基因的失活与原发性卵巢癌的发生有关。
Objective To study the expression of p15 mRNA and protein in primary ovarian carcinoma, and to define the role of p15 gene in the generation and development of the disease. Methods By using reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemistry assays, expressions of p15 mRNA and protein were detected in 48 primary ovarian carcinomas and 15 normal ovarian tissues. Results Significant differences existed in the expression of p15 mRNA and protein between the two groups(χ 2=4.16,5.11;P<0.05). The expressions of p15 mRNA and protein correlated with the histological types (χ 2=5.59,7.30;P<0.05), but not with FIGO stage and histological grade(χ 2=1.13-2.09,P>0.05). Conclusion In primary ovarian carcinoma, the expressions of p15 mRNA and its protein are abnormal, which suggestive of that inactivation of p15 gene is associated with oncogenesis
