bFGF对骨骼肌肌卫星细胞增殖的影响—PCNA免疫组化法的实验研究 被引量:5

摘要 目的 :试图用致中胚层来源细胞分裂剂—碱性成纤维细胞生长因子 (basicfibroblastgrowthfactor ,bFGF)使正常成熟骨骼肌肌卫星细胞从静止状态变成增殖活动。方法 :用 2 1只大白鼠腓肠肌为实验材料 ,随机分 3组 ,每组各 7只 ,A组肌肉内注入bFGF ;B组肌肉内注入生理盐水 ;C组肌肉内不注任何物质。连注 14d后 ,取各组腓肠肌制成HE染色和增殖细胞核抗原 (proliferatecellnucleusantigen ,PCNA)染色切片 ,最后光镜下观察和对观察相关资料做统计学处理。肌卫星细胞核 ,HE染色为蓝色 ,PCNA染色阳性为棕黄色。结果 :细胞核数量 ,A组较B、C两组多 ;B组较C组多 ;C组肌肉内几乎未见有PCNA阳性细胞核存在。结论 :bFGF可使骨骼肌的肌卫星细胞 ,由静止变成增殖状态 ,为解决成体干细胞快速增殖这一难题 。 Objective: To change the immovable state which normal muscle satellite cells are into the proliferative state by adopting basic fibroblast growth factor (bFGF). Methods: The gastrocnemius in 21 white big mice were adopted and were divided into A, B, C group. There are 7 mice in every group. A group: bFGF was injected into the gastrocnemius. B group: 0.9saline was injected into. C group is control group and nothing was injected into. Cut off materials after 14 days, they are respectively made into optical routinely and stained H.E, PCNA and observed and tested under the light microscope .And the material of statitus was mamaged. The nuclear of the muscle satellite cell are stained blue by H.E and stained brown-yellow by PCNA. Results: In A group, the guantities of the nuclear of the muscle satellite cells are most of the three groups, and then B group is more than C group. There isn′ t the nuclear of PCNA (+) in C group. Conclusion: bFGF can make the muscle satellite cell change the immovable state into proliferative state. This will provide the guidance for solving the mature stem cells rapidly into proliferative problem.
