摘要
目的 建立高效液相色谱 电化学检测法 (HPLC EC)测定人血浆中地红霉素的活性代谢物红霉素胺浓度。方法 以吲达帕胺为内标 ,血浆经碱化采用二氯甲烷提取 ,色谱柱为KromasilC18,流动相甲醇 :乙腈 :pH 7.8磷酸盐缓冲液为 13∶4 0∶4 7,流速为 0 .7mL/min ,电化学检测器工作电压为 +0 .9V。结果 红霉素胺血浆浓度在13.8~ 4 6 0ng/mL内具有良好线性关系 ,方法回收率 99.2 %~ 10 5 .4 % ,日内、日间相对标准差 (RSD)均 <10 %。结论 本方法简便、准确、灵敏、重现性好 ,可用于地红霉素的血药浓度监测及药代动力学研究。
Objective To establish a HPLC method for the determination of erythromycylamine in human plasma by electrochemical detection. Methods The internal standard was indapamine, and the samples were extracted by dichloromethane after the plasma was basified. The column was KromasilC 18 , and the mobile phase consisted of CH 3OH∶CH 3CN∶BUFFER (13∶40∶47). The applied potential of the amperometric detection was set at +0.9 V , and the flow was 0.7 mL/min . Results The method was linear in the range of 13.8 ~460 ng/mL. The RSD of within-day and between-day were less than 10%. The method recovery was 99.2% ~ 105.4% . Conclusion The method is convenient, accurate, sensitive, reliable, and effective for monitoring plasma concentration and pharmacokinetics studies of dirithromycin.
