摘要
目的探讨匹多莫德对小儿辅助T细胞亚群功能的影响。方法常规分离新生儿脐血单个核细胞(CBMC)和毛细支气管炎患儿外周血单个核细胞(PBMC),加入匹多莫德,ELISA检测培养上清中的IL鄄4、IFN鄄γ、IL鄄10和IL鄄12水平。结果匹多莫德可显著下调PHA诱导的CBMCIL鄄4分泌,上调LPS诱导的IL鄄12p40和IL鄄12p70的分泌;匹多莫德诱导毛细支气管炎患儿PBMC产生IFN鄄γ,恢复患儿体内相对亢进的Th2细胞功能,单独匹多莫德诱导毛细支气管炎患儿PBMC产生较高水平的IL鄄12p70(135±44pg/ml,n=9),较低水平的IL鄄10(52±16pg/ml,n=9)。结论匹多莫德具备调节Th1/Th2细胞功能平衡的能力,其机制可能与匹多莫德作用于抗原提呈细胞,刺激IL鄄12产生有关。
Objective: To explore the effects of pidotimod on T cell subsets of children. Method: Neonatal cord blood mononuclear cells (CBMC) and peripheral mononuclear cells (PBMC) of children with bronchiolitis were isolated to investigate the effects of pidotimod. The level of IL-4, IFN-γ, IL-10和IL-12 in supernatant were detected by ELISA. Results: Pidotimod significantly down-regulated IL-4 secretion of CBMC induced by PHA, and up-regulated IL-12 p40 and p70 production induced by LPS. Moreover, pidotimod increased IFN-γ production from PBMC of children with bronchiolitis. Pidotimod alone could induce relatively higher IL-12 p70 (135±44 pg/ml, n=9) and lower IL-10 (52±16 pg/ml, n=9) production. Conclusion: Pidotimod could regulate the balance of Th1/Th2. The mechanism of Pidotimod might relate to its impact on antigen presenting cell to induce IL-12.
