盐酸文拉法辛的合成

Synthesis of Venlafaxine Hydrochloride

用对甲氧基苯乙腈和环已酮在氢化钾/甲苯中进行缩合反应得到a-(1-羟基环己基)-对甲氧基苯乙腈,再经Red-Al或NiCl2/NaBH4还原、甲基化等反应制得盐酸文拉法辛,总收率为50%～62%。

Venlafaxine hydrochloride was synthesized from 4-methoxybenzyl cyanide and cyclohexanone bycondensation in presence of KH in toluene to give a-(1-hydroxycyclohexyl)-4-methoxybenzyl cyanide, which subjectedto reduction by Red-Al or NiCl2/NaBH4 and subsequent methylation with an overall yield of 50%～62%.
