摘要
用沉淀法分离大鼠血清高密度脂蛋白(HDL),SephadexG—200层析纯化;将脱脂的HDL(ApoHDL)通过SephadexG—200柱层析得到单一的载脂蛋白A-I(ApoA-1)。SDS-聚丙烯酰胺凝胶电泳(SDS—PAGE)测定该ApoA-Ⅰ的表观分子量为27000道尔顿,其免疫学特征及化学组成分析结果与文献报道一致。同时用超速离心法分离制备出纯化的大鼠血清低密度脂蛋白(LDL)。
ApoA-Ⅰ is the major apolipoprotein in the precipitate by the sodium edxtran sulfate 500 000—Mn^(2+) reagent (DS50—Mn^(2+)) from rat serum. HDL was purified from the crude precipitate HDL by sephadex G—200 chromatography and then delipidated by the ethanol—ether procedure. The ApoA-Ⅰ was then chromatographied on the sephadex G—200 column eluted by 0.01M Tris-HCL buffer. The rat serum LDL is isolated by ultracentrifugation. The apparant molecular weight of the purified rat ApoA-Ⅰ determined on SDSPAGE, was 27000 dalton. Both the ApoA-Ⅰ and LDL's immunological properties and chemical compositions were in accordance with the previous reports.
