α-乙酰乳酸的合成及α-乙酰乳酸脱羧酶产生菌的筛选与鉴定 被引量:2

摘要 利用有机反应合成α 乙酰氧基 α 甲基 乙酰乙酸乙酯,后者经NaOH水解生成的α 乙酰乳酸,可以用作测定α 乙酰乳酸脱羧酶(简称ALDC)活力的底物.通过富集,从土壤和水体中分离出100株产芽孢的菌株,其中获得VP反应为阳性的37株芽孢杆菌,通过对ALDC活力的反复测定,得到7株有较高酶活性的菌株,并根据其生理生化的特征进行了初步的鉴定,为进一步开发和利用α 乙酰乳酸脱羧酶奠定了基础. α-acetooxy-α-methyl-acetoacetic acetate was synthesizd by organic reactions. By the hydrolysis with NaOH, it could produce α- acetolactic acid, which was used as the substrate to determine the activity of α-acetolactate decarboxylase(ALDC). 100 strains of spore-forming bacteria were isolated from the soil and the water. 37 strains of them were positive in VP. The activity of α-acetolactate decarboxylase was repeatedly determined by using two kinds of substrate. Seven strains with higher activity of ALDC were obtained, and they were identified with morphophological, physiological, bio-chemical analyses. The study is of benefit to the improvement and the application of ALDC in industry.
