复方861对人肝星状细胞α-平滑肌肌动蛋白mRNA表达的调节作用 被引量:5

摘要 目的 研究复方861对人肝星状细胞(LX-2)中α-平滑肌肌动蛋白(α-SMA)基因表达的调节作用。方法应用基于SYBR-Green I的实时定量荧光PCR方法,研究复方861分别作用24、48、72 h后,LX-2细胞中α-SMA mRNA含量的变化情况。结果 复方861在48和72 h可以显著降低LX-2细胞中α-SMA mRNA含量。结论 复方861可抑制LX-2细胞中α-SMA基因的表达,提示其抗肝纤维化作用可能与其抑制LX-2细胞的活化有关。 Object To investigate the effects of Compound 861 (Cpd861) on expression of α-smooth muscle actin (α-SMA) in human hepatic stellate cells (LX-2 cells). Methods Real-time quantitative PCR method using SYBR Green I technology was taken to measure the effects of Cpd861 on expression of α-SMA mRNA in LX-2 cells treated by Cpd861 for 24, 48 and 72 h. Results The expression levels of α-SMA mRNA decreased significantly in LX-2 cells exposed to Cpd861 for 48 h (P<0. 05) or 72 h (P< 0. 01). Conclusion Cpd861 can reduce the expression levels of α-SMA mRNA in LX-2 cells.
