血管内皮生长因子(VEGF)基因转染促进成骨细胞成骨活性的实验研究 被引量:8

Experimental study of effects of pBLAST49 mVEGF gene transfection on function of osteoblast cells

摘要 目的了解外源性血管内皮生长因子(VEGF)基因表达对成骨细胞成骨活性的影响。方法利用阳离子脂质体Lipofectamine转染将pBLAST49-mVEGF基因导入体外培养的成骨细胞,与对照组比较,观察外源性VEGF在成骨细胞内的表达,及其对成骨细胞合成分泌骨钙素、Ⅰ型胶原的影响。结果pBLAST49-mVEGF基因转染组第1~5代细胞合成分泌骨钙素浓度和Ⅰ型胶原的表达均明显高于对照组,二者间比较差异具有显著性(P<0.05)。结论pBLAST49-mVEGF基因转染能促进成骨细胞生成骨钙素和合成Ⅰ型胶原的能力。 Objective To understand the effects of extra VEGF gene expression on the maturation and function of osteoblast cells. Methods The extra VEGF gene was induced into osteoblast cells in vitro by lipofectamine. The expression of extra VEGF gene, and its effect on the synthesis of type Ⅰ collagen and secretion of osteocalcium of osteoblast cells were observed. Results The concentration of osteocalcium and expression of type Ⅰ collagen in osteoblast cells of the 1st to 5th generations in the pBLAST49 mVEGF gene transfer group were higher than those in the control group. Significant statistical difference existed (P< 0.05). Conclusion Extra pBLAST49 mVEGF gene transfer can improve the synthesis of type Ⅰ collagen and secretion of osteocalcium in osteoblast cells.
