摘要
目的 :研究丁酸盐对人类珠蛋白基因表达的影响。方法 :选用K5 6 2细胞为体外模型 ,采用联苯胺染色和RT -PCR技术 ,比较丁酸盐诱导前后联苯胺染色阳性率和 7种珠蛋白基因mRNA改变。结果 :丁酸盐诱导后联苯胺染色阳性细胞率 (BZ % )增加 3~ 6倍 ;Gγ -mRNA增加 2 .2 6± 0 .2 2倍 (P <0 .0 5 ) ,α、ζ、ε、δ、Aγ -mRNA均有增加但增加不显著 (P >0 .0 5 ) ;诱导前后K5 6 2细胞均未检测到 β -RNA。结论 :丁酸盐可诱导珠蛋白基因表达并选择性地刺激γ基因转录增加 ,尤其是Gγ ;
Objective:To investigate the effects of butyrate (BA) on the expression of human globin genes. [WT5HZ]Methods: K562 cells,used as a in vitro model system, were stained with benzidine to detect hemoglobin production. Comparison of the percentage of benzidine-positive cells(BZ%) was performed in K562 cells of untreated and treated with different concentration of butyrate. The technique of RT-PCR was employed to determine the changes of seven kinds of globin mRNA in untreated and butyrate-treated K562 cells. [WT5HZ] Results:The BZ% was increased 3 to 6 folds, Gγ-mRNA was increased 2.26±0.22 fold( P <0.05), and α,ε,ζ,δ-mRNA were increased insignificantly( P >0.05) after treatment with BA.The β-mRNA was not found in K562 cells even after 4 to 5 days drug treatment. [WT5HZ]Conclusion:Butyrate can induce the expression of globin genes and augment of γ gene transcription in K562 cells, especially in Gγ-mRNA.The induction of butyrate is similar to that of hydroxyurea which used as a positive control.
