摘要
目的 研究人胶质瘤组织中猴多瘤病毒SV4 0及人多瘤病毒 (JCV和BKV)的表达情况 ,阐明多瘤病毒与人胶质瘤病因的相关性 ,为胶质瘤的预防和治疗提供理论依据。方法 采用聚合酶链反应 (PCR)方法检测 94例胶质瘤和 18例正常脑组织中SV4 0、JCV和BKV的基因片段 ,用免疫组化检测SV4 0大肿瘤抗原 (LT)的表达。结果 在胶质瘤中SV4 0、JCV和BKV的检测阳性率分别为5 7.4 5 %、4 8.94 %和 19.15 % ;4 6 .8%的胶质瘤表达LT蛋白。胶质瘤和正常脑组织间SV4 0DNA、JCVDNA及SV4 0LT蛋白检测阳性率差异有显著性 ;不同分级的星形细胞肿瘤间SV4 0LT蛋白阳性率差异无显著性 ;PCR和免疫组化用于检测SV4 0具有相关性 ,差异无显著性 (P >0 .0 5 )。结论 SV4 0与人胶质瘤的发生密切相关 ;JCV可能是胶质瘤发生的诱因之一 ;免疫组化与PCR可作为临床对SV4
Objective In order to provide new insights for the prevention and therapy of gliomas, the research tested the possibility that monkey polyomavirus Simian virus 40(SV40),human polyomavirus JC virus (JCV )and BK virus (BKV) were associated with human gliomas.Methods Polymerase chain reaction (PCR) was used to detect SV40, JCV and BKV in 94 glioma samples and 18 normal brain tissue samples. The expression of SV40 large tumor antigen (LT) was examined by immunohistochemistry.Results In glioma, the positive rates of SV40, JCV and BKV were 57.45%, 48.94% and 19.15% respectively. There were significant differences in SV40 DNA, JCV DNA and SV40 LT( P <0.05) between gliomas and normal tissues. No significant difference was found among different grades of astrocytomas. A correlation was observed between PCR and immunohistochemistry in the detection of SV40( P >0.05). But no difference was found between two methods.Conclusion SV40 is closely related to gliomas and plays an etiologic role in the development of gliomas. JCV is related to gliomas and may be involved in the development of gliomas. Both PCR and immunohistochemistry are reliable methods to detect SV40 at clinic.
