Regulation of glioma invasion by Insulin-like Growth Factor Binding Protein 2(IGFBP2) and its binding partner IIp45

摘要 经过对不同级别胶质瘤的基因表达谱研究发现,在高级别胶质瘤(胶质母细胞瘤和多形性胶质母细胞瘤)中胰岛素样生长因子结合蛋白2 (IGFBP2)呈过度表达,随之进行的微阵列分析表明80%的多形性胶质母细胞瘤过表达IGFBP2,这是目前为止发现的多形性胶质母细胞瘤最普遍的分子生物学改变。功能学研究表明:IGFBP2能促进胶质瘤的侵袭性。通过采用酵母双杂交技术,确定了编码IGFBP2结合位点的IIp45基因,IIp45蛋白的作用为抑制胶质瘤的侵袭性。总之,基因功能学研究表明IGFBP2和IIp45为一对作用相反的胶质瘤侵袭性调节蛋白。 Gene expression profiling studies with glioma tissues of different grades led us to the discovery that insulin-like growth factor binding protein 2 (IGFBP2) is overexpressed in the advanced stage of glioma-glioblastoma multiforme or GBM. Subsequent studies including tissue microarray analysis revealed the IGFBP2 overexpression is present in 80% of GBM and by far is the most frequent molecular event uniquely observed in GBM. Functional studies showed that IGFBP2 is an enhancer of glioma invasion. Using yeast two-hybrid technique, we have identified a novel gene IIp45 that encodes a binding partner of IGFBP2. The function of IIp45 is inhibition of invasion. Thus, our functional genomic studies have characterized IGFBP2 and IIp45 as a pair of interacting proteins with opposing functions in glioma invasion regulation.
