摘要
目的 :探讨局部应用白细胞介素 12 (IL 12 )对于小鼠皮下肿瘤的治疗作用。方法 :小鼠右侧腹部皮下注射 2x10 5个Panc 0 2细胞建立皮下肿瘤模型。皮下肿瘤被检测 2 4d ,通过测量的皮下肿瘤的两个直径以计算出肿瘤大小 (mm2 )。分为IL 12治疗组和对照组 ,每组 6只小鼠。从第 3~ 7天 ,IL 2治疗组 ,每天在肿瘤周围注射 0 .1μgIL 12 ,对照组同时同部注射同量平衡盐水。 结果 :与对照组相比 ,IL 12治疗组从第 7天开始Panc 0 2肿瘤的生长出现受到抑制。第 10天 ,IL 12治疗组 [(13.8± 8.7)mm2 ]与对照组 [(36 .4± 8.5 )mm2 ],(P <0 .0 5 )之间的差异有统计学意义 (P <0 .0 5 )。第 2 4天 ,两组之间的差异更加明显 [(6 6 .5± 5 0 .4 )mm2 VS (2 6 4 .4± 4 6 .4 )mm2 ,P <0 .0 1]。结论 :肿瘤周围注射IL 12明显抑制Panc 0 2皮下肿瘤的生长 。
Objective:To investigate the anti-tumor effect of IL-12 in treatment of murine subcutaneous tumor.Methods:2×10^(5) Panc-02 cells were injected subcutaneously into the murine right flank to establish the subcutaneous tumor model. The growth of the subcutaneous tumors was monitored in 24 days, and tumor size (square millimeters) was shown by the product of the two perpendicular diameters of each tumor.The established palpable tumors were observed on day 3. From day 3 to 7,0.1 μg IL-12 was peritumorally administered.Results:From day 7, the growth of Panc-02 tumors in IL-12 treatment group was slower when compared with control group. The tumor growth in IL-12 treatment group (13.75±8.66 mm^(2)) was significantinhibited on day 10 compared with that in control group(36.40±8.50 mm^(2), P<0.05). The difference between these two groups was more significant on day 24 (66.50±50.36 mm^(2), 264.40±46.35 mm^(2), P<0.01). Conclusion:Peritumoral administration of IL-12 was effective to inhibited subcutaneous tumor growth, and would be effective in clinical application.
