摘要 以士的宁为工具药,研究了环境温度、给药剂量、动物体重和给药时辰等因素对牛蛙药效的影响.结果表明,1)环境温度对药效有显著影响.在一定范围内,随着温度的升高,牛蛙出现惊厥的潜伏期越来越短;但不同温度范围对其的影响程度不一样,在0~10 ℃,30~35 ℃,士的宁药效对温度变化极为敏感.2)给药剂量对药效也有显著影响.在每10 g剂量0.01~0.20 mL范围内,随着剂量的增加,牛蛙出现惊厥的潜伏期也越来越短,而不同剂量段对其药效的影响是不同的.在蛙体上,士的宁也同样表现出安全范围较窄的特点.3)在42.8~203.5 g体重范围内,体重对药效有一定的影响.4)给药时辰对药效也有影响,并表现出昼夜节律性.在12:00左右,牛蛙对士的宁的敏感性大大降低.5)在四个影响因素当中,环境温度和给药剂量是影响牛蛙药效的主要因素. The strychnine was used as instrument to study the effects of ambient temperature,dosage,body weight and dosing time on drug action in Rana catesbeiana.The results indicated:1)The effect of ambient temperature on the action was remarkable.In the specific range,the higher the ambient temperature was,the shorter the latent period of stimulation was.But the degree of effect was diverse between different ranges.The action varied very sensitively to the temperature change from 0 ℃ to 10 ℃ and from 30 ℃ to 35 ℃.2)The general change of effect of dosage was similar to that of temperature,and the strychnine in bullfrog showed the same characteristics of narrow margin of safety as in other animals.3)In the scope of body weight from 42.8 g to 203.5 g,the effect was found,but not as remarkable as temperature was.4)The influence of dosing time was also found.It represented a significant circadian rhythm when strychnine was given to bullfrog at different time of the day.And the sensitivity of bullfrog to strychnine was lower around twelve oclock.5)Among ambient temperature,dosage,body weight and dosing time, the first two are the main factors influencing the drug action.
