摘要
以4个辣椒品种茎尖为外植体,在MS+4mg/LBA+1mg/LIAA+4mg/LAgNO3培养基上诱导不定芽分化,在MS+2mg/LBA+0.1mg/LIAA,MS+0.5mg/LBA+0.1mg/LIAA诱导不定芽茎伸长.结果,平均不定芽诱导率达85%,平均不定芽茎伸长率可达110%,再生植株频率达75%左右.初步实验结果表明:与子叶及下胚轴培养相比,茎尖培养具有成苗率高、基因型不敏感、再生周期短等优势,是解决辣椒组织培养不定芽茎伸长困难,提高再生植株诱导频率的有效途径.
Shoot-tips of four pepper varieties (Capsicum annuum L.) were used as explants in the present experiment. The explants were cultured on MS+(4 mg/L) BA+(1 mg/L) IAA+(4 mg/L) (AgNO_3) to induce adventitious buds. Subsequently, the buds were cultured on MS+(2 mg/L) BA+(0.1 mg/L) IAA, MS+(0.5 mg/L) BA+(0.1 mg/L) IAA to induce adventitious stem and elongation. As a result, the average rate of buds differentiation was over 85%, the average rate of buds reached 110%, and the average rate of regeneration was 75%. Compared to the regeneration system of coty-ledon explants, shoot-tip regeneration system exhibited many advantages such as high rate of regene-ration, low sensitivity of genotype, the short cycle of regeneration, etc. It is an effective and quick (regeneration) system.
