鼠伤寒杆菌外膜蛋白的提取和分析鉴定

Extraction and analytical identifcation of outer membrane proteins of Salmonella typhimurium

采用超声波破碎、Tritonx-100处理和超速离心的方法提取鼠伤寒杆菌外膜蛋白(OMP)。经电镜和SDS-PAGE分析鉴定,鼠伤寒杆菌的主要OMP带位于36～41kD之间,应用Western blot分折抗-OMP的免疫血清,显示血清抗体所识别的主要是36kD蛋白带。试验证明,用该种方法提取OMP是可行的,36kD蛋白可能是OMP中主要免疫原。

OMPs of S. typhimurium were extracted with the method described by Schnaitman, and wereanalysed with SDS-PAGE. The 36-41 kD protein bands were the major OMP bands. Immunologicalserum of OMP was assayed with Western blot. The results showed that 36 kD OMP from variousstrains could be distinguished. The 36 kD proteins may be a major immunogen of OMP.
