新的双试剂自动分析法测定血清铜:5-Br-PADCAP直接光度法

A new automatic method for analysis of copper in serum with 2 reagents:5-Br-PADCAP direct spectrophotometry

〔目的〕建立简便、灵敏的双试剂自动化分析法测定血清铜。〔方法〕在表面活性剂存在下 ,用 2 (5 溴 2 吡啶偶氮 ) 5 [(N ,N 二羧基甲基 )氨基 ]苯酚 (5 -Br -PADCAP)为显色剂测定血清铜。〔结果〕该法线性范围 0～ 65 μmol/L ,平均回收率为 10 0 .7% ,批内变异系数 (CV)和批间变异系数分别为 0 .0 2 4、0 .0 3 6,与原子吸收分光光度法比较具有良好的相关性 ,线性回归方程和相关系数分别为Y =1.10 47X -0 .15 3 8,r =0 .9897。 5 4例健康人血清铜含量为 8.2 9～ 2 2 .45 μmol/L(x± 2s)。〔结论〕用 5 Br PADCAP双试剂自动化分析法测定血清铜方法简便、灵敏可靠 。

〔Objective〕To establish a simple,sensitive automatic method for determination of copper in serum with 2 reagents.〔Methods〕 The serum copper was detected with 5-Br-PADCAP with the presence of surfactant.〔Results〕 The linear range was 0～65 mmol/L,the average recovery was 100.7%,the within- and between-run CV was 0.024 and 0.036,respectively,for the method.The comparison of this method (Y) with AAS (X) resulted in the regression equation:Y=1.1047X-0.1538, r =0.9897, P >0.05.The reference interval was 8.29～22.45 mmol/L (n=54).〔Conclusions〕 This method is simple,sensitive and suitable for clinical application.
