保健食品中大豆异黄酮的测定方法研究 被引量:15

摘要 〔目的〕检测保健食品中黄豆甙、黄豆甙元、染料木甙和金雀异黄素 ,建立保健食品中四种大豆异黄酮的二极管阵列检测器高效液相色谱法。〔方法〕样品用甲醇 +水 (80 + 2 0 ,v/v)超声提取 3 0min ,定容后过 0 .45 μm滤膜 ,上 2 5 0×4.6mm ,粒径 10 μm的C18柱 ,以甲醇 + 0 .0 5 0mol/L ,pH4.6醋酸铵 (4 6+ 5 4,v/v)作流动相 ,二极管阵列检测器测定。测定时间只需 2 0min左右 ,黄豆甙 ,染料木甙 ,黄豆甙元和金雀异黄素可得到良好分离。〔结果〕黄豆甙 ,染料木甙 ,黄豆甙元和金雀异黄素的加标回收率分别为 90 .4%~ 10 2 % ,88.9%~ 10 2 % ,93 .0 %~ 10 2 % ,89.3 %~ 10 4% ,加标回收相对标准偏差均小于 6% ,最底检出浓度均为 0 .10 μg/ml。 〔Objective〕To develop a method based on HPLC-PDA for determination of 4 soy isoflavones in health foods.〔Methods〕 After sonicated for 30 min and filtered thromgh the 0.45 um filter,the samples were separated on a C18 reversed-phase column using methanol:0.050 mol/L ammonium acetate pH4.6 as the mobile phase at a velocity of 1.2 ml/min and detected by photodiode-array detector at 210nm-400nm.Good separation of the daidzin genistin,daidzein and genistein was achieved in about 20 min.〔Results〕 The recovery of daidzin genistin,daidzein and genistein was 90.4%~102%,88.9%~102%,93.0%~102% and 89.3%~104%,respectively,and the variation coefficient was below 6% for all.The minimal detecting concentration of the 4 soy isoflavones was 0.10 mg/ml.〔Conclusions〕 This method is suitable for the determination of soy isoflavones in health foods.
