香菇C_(91-3)菌丝发酵液蛋白抑制小鼠宫颈癌细胞株U14生长及诱导凋亡的实验研究 被引量:21

摘要 目的研究香菇C91-3菌丝发酵液蛋白LFP91-3对小鼠腹水型宫颈癌细胞株(U14)的作用。方法应用不同剂量(50、100、150μg)的LFP91-3对U14所致小鼠腹水型宫颈癌模型进行治疗,观察荷瘤小鼠的生存期;并用MTT法LFP91-3浓度5、10、15μgml和流式细胞仪对经LFP91-3作用的U14进行观察和检测。结果LFP91-3能明显延长U14所致荷瘤小鼠的生存期。对体外培养的U14有直接杀伤作用,抑制率随LFP91-3浓度的增加和作用时间的延长而升高。流式细胞仪检测结果显示LFP91-3阻滞U14于细胞周期的分裂中期。结论LFP91-3能抑制小鼠宫颈癌细胞株的生长并诱导其凋亡。 Objective To investigate the effect of Lentinus dodes C91-3 my celium fermentative liquid protein (LFP91-3) on the growth of mouse carcinoma ce ll of cervix uteri line U14. Methods Survival time of U14 tumor-bearing mice wa s observed after the animals were treated with LFP91-3 of different concentratio ns (50ìgml, 100ìgml, 150ìgml). LFP91-3 induced apoptosis of U14 cells w ere detected by MTT assay and flow cytometry after tumor cells were treated with different concentrations of LFP91-3 (5ìgml?10ìgml?15ìgml). Results Th e survival time of tumor-bearing mice were significantly prolonged with the trea tment of LFP91-3. The growth of U14 cells was significantly inhibited by LFP91-3 and the inhibitory rate was depended on the LFP91-3 concentrations and treatmen t time. Flow cytometry showed the typical cell apoptosis cell cycle stopped at S phase. Conclusion The LFP91-3 can significantly inhibit cell proliferation a nd induce the apoptosis of mouse carcinoma U14 cells of cervix uteri.
