摘要
目的 :研究肺癌端粒酶活性和临床特征的相关性 ,评价端粒酶作为肺癌诊断标记物的可行性 ,初步探讨吸烟和端粒酶激活在肺癌发病机制中的作用。方法 :组织标本取自 2 0 0 0年 6月~ 2 0 0 1年 1月华西医大附一院胸外科住院手术病人 ,其中原发性肺癌 2 7例 ,结核瘤 2例 ,炎性假瘤 1例。用 TRAP- EL ISA法检测端粒酶活性。结果 :2 7例组织病理学确诊的肺癌有 81.4 8% (2 2 / 2 7)端粒酶活性检测阳性。在不同组织病理学分型中 ,阳性率由高到低依次为 SCL C (小细胞肺癌 )、鳞癌、复合性腺鳞癌、细支气管肺泡腺癌、腺癌。肺癌组织端粒酶活性明显高于癌旁 (P=0 .0 0 0 )和良性组织 (P=0 .0 13)。结论 :端粒酶是一种较理想的诊断性肿瘤标志物。吸烟可能通过端粒酶激活途径促发鳞癌。
Objective:Lung cancer is a typical disease related to individual heredity susceptibility,environmental-vocational factors and life styles.More than half of them is mainly related to environmental and vocational exposure.This research work is to investigate the activities of telomerase in human lung cancer tissues and the correlation of telomerase activity with the clinical features,to evaluate the significance of this diagnostic tumor marker and to explore the role of smoking and telomerase in carcinogenesis.Methods:Operation specimens came from hospitalized patients from June,2000 to January,2001 in department of chest surgey of West China University of Medical Sciences,including 27 originated lung cancers,2 tuberculomas and 1 inflammatory pseudotumor.Telomerase activity was detected using telomeric repeat amplification protocal enzyme-linked immunoadsorbent assay(TRAP-ELISA).We analysis the correlation of telomerase activity with clinical features in lung cancer.Results:Telomerase activity was detectable in 81.48% of 27lung tumour tissues,but none of 3 benign lung tissues.The positive rate of telomerase activity is decreasing in the order of SCLC,squamous carcinoma,adenosquamous carcinoma,bronchiole alveolar carcinoma and adenocarcinoma.The telomerase activity of lung tumor tissues is significantly higher than that of paired adjacent tissues(P=0.000) and benign lung tissues(P=0.014).Conclusion:The current results shows that telomerase is a rather novel diagnostic tumor marker of pulmonary carcinoma.Smoking may promote squamous carcinogenesis by telomerase activation pathway.
