胰腺癌组织ER mRNA与PS2表达的相关性研究

Correlative study on the expression of ER mRNA and presenilin 2in the pancreatic carcinoma tissues

目的 :研究胰腺癌组织ERmRNA和PS2表达特征、相互关系及其临床病理意义。方法 :35例胰腺癌手术切除标本经 1 0 %中性福尔马林固定后常规制作石蜡包埋切片 ,ERmRNA染色为原位杂交染色法 ,PS2染色为常规ABC免疫组化法。结果 :35例胰腺癌PS2阳性 1 6例 (4 5 .7% ) ,其评分值为 1 .82± 1 .34;ERmRNA阳性 1 8例(5 1 .4 % ) ,其评分值为 2 .0 8± 1 .4 6。PS2和ERmRNA表达阳性率及其评分之间均存在密切正相关 (γ =0 .6 4 ,P <0 .0 1 )。高分化腺癌PS2和ERmRNA表达阳性率及其评分明显高于中、低分化腺癌 (P <0 .0 5或P <0 .0 1 ) ,无转移癌两者阳性率及其评分高于转移癌 (P >0 .0 5 )。结论 :PS2和ERmRNA表达与胰腺癌生物学行为及其雌激素依赖性存在密切关系 。

Objective To study the expression of ER mRNA,presenilin 2 (PS2) and their relationship and clinicopathological significance in the pancreatic carcinoma. Methods The assay of ER mRNA was detected by in situ hybridization technique and the expression of PS2 was completed by immunohistochemical method of avidin biotin complex in the 10% neutral formalin fixed and routinely paraffin embedded sections of specimen with pancreatic carcinoma.Results The positive rate and the score of PS2 were 45.7% and 1.82±1.34, also those of ER mRNA were 51.4% and 2.08±1.46 in 35 patients with pancreatic carcinoma .The highly positive correlations were found among the positive rate and the score of ER mRNA and PS2. The positive rate and the score were significantly higher in the cases of well differentiated adenocarinoma without metastasis than those in the ones of middle or poorly differentiated adenocarinoma with metastasis.Conclusion The expression of ER mRNA or PS2 might reflect the biological behaviors and hormone dependent status of pancreatic carcinoma. The assay of ER mRNA and meanwhile the detection of PS2 might have important clinical values for better understanding sex hormone dependence and raising the therapeutic effect of clinical endocrine therapy of the pancreatic carcinoma.
